pH experiment: from measurements to the observed maximum

The pH tab builds a measurement plan and a curve from your data. It finds the largest mean among tested points; it does not predict a new pH optimum or automatically select a buffer.

Record conditions and units

Specify protein, temperature, method, buffer, substrate, batch and activity units. Comparing points requires clear conditions. Account for buffer changes between points: the table cannot separate pH effects from every other experimental factor.

The beta accepts a range within 0–14, steps of 0.1–3 and two to six replicates, with up to 40 points and 180 measurement fields. These are interface limits, not recommendations for experiments under extreme conditions.

Fill the table manually or through CSV

Creating a plan gives pH–replicate–activity rows. Download, fill and reimport the table, or enter values in the browser. A blank cell means a missing measurement; enter the number 0 for a measured zero.

Minimum CSV columns: ph,replicate,value. Replicate numbers must match the plan. Any conditions included in the file must match the current series. Unknown points, duplicate rows and invalid numbers are rejected.

What is calculated?

For each point, the service uses only entered numbers to calculate the arithmetic mean and sample standard deviation, SD. For 80, 82 and 84, the mean is 82 and SD is 2. SD is not calculated for a single measurement. Missing values are not replaced by zeros.

The chart connects measured means for readability; vertical bars show replicate SD. They are not confidence intervals or a model of a chemical relationship. Actual activity between tested points is not calculated.

Interpret the maximum and plan the next series

If two pH points share the highest mean, both are shown. A maximum at the plan boundary triggers a notice to test neighboring values. Missing replicates are also flagged; the conclusion may be preliminary.

Use a smaller step around the observed maximum in the next series and check reproducibility. This is the experimenter’s decision, not an automatically executed protocol. Decide in advance whether you compare absolute activity, relative activity or stability after treatment.

Save and transfer results

Measurements are saved locally in this browser. Use CSV for tables and JSON to preserve the series state and manually restore it on another device. Importing JSON recalculates summaries from measurements; precomputed conclusions in the file are not trusted.

There is no automatic synchronization or model training on these data. A laboratory account is a separate development stage. Clearly marked DEMO data are available for a first trial and stored separately from your working series.
